摘  要:目的了解宁夏地区绵羊脑组织博尔纳病病毒(Borna disease virus,BDV)的自然感染情况,并分析其与德国马源性标准病毒株He/80的同源性。方法应用荧光定量巢式逆转录PCR(FQ-nRT-PCR)和RT-PCR分别检测宁夏地区163例绵羊脑组织中BDV p24、BDV p40基因片段,并对BDV p24阳性扩增产物进行测序,与国外标准病毒株进行比较。结果 163例绵羊脑组织中6例BDV p24、BDV p40均阳性,阳性率为3.68%(6/163)。测序后经基因序列比对,本实验检测阳性BDV p24片段的核苷酸序列与He/80同源性为100%。结论宁夏地区绵羊脑组织中可能存在动物源性BDV的隐性感染,阳性BDV p24核苷酸序列与He/80具有高度同源性。Objective To investigate BDV naturally infected in brain tissue of sheep in Ningxia,and to explore the homology with the German horse-borne standard strain He/80.Methods BDV p24 gene and BDV p40 in brain tissue of 163 sheep cases in Ningxia were detected with Fluorescence quantitative nested reverse transcriptase PCR(FQ-nRT-PCR) and RT-PCR detection.The BDV p24 positive samples were sequenced and compared with the foreign standard virus strains.Results The positive rate was 3.68%(6/163) in brain tissue of sheep.after sequence comparison,BDV p24 fragment of the nucleotide sequence and He/80 showed 100% homology.Conclusion The brain tissue of sheep of Ningxia may exist latent infection of BDV and the positive BDV p24 nucleotide sequence is highly homologous with He/80.
