摘 要:目的:探讨新生鼠短期暴露低剂量毒死蜱(chlorpyrifos,CPF)对海马星型胶质细胞和远期神经行为的影响。方法:制作出生后11-14d Sprague-Dawley大鼠低剂量CPF暴露模型。在出生后15d、20d、30d、60d,观察有无急性有机磷农药中毒症状、体重增长情况、脑组织外观、脑系数和脑含水量;免疫组织化学方法检测海马星形胶质细胞胶质纤维酸性蛋白表达。青春期和成年期用情绪水平量表检测其整体情绪水平;高架十字迷宫实验评估焦虑水平。结果:低剂量CPF暴露后未见急性有机磷农药中毒症状,体重增长、脑组织外观、脑系数、脑含水量均未见异常;但海马GFAP的表达上调(P<0.05);十字迷宫实验中在开放臂停留的时间比率减少(P<0.01),并降低整体情绪水平(P<0.01)。结论:在生命早期无近期中毒症状和脑外观改变的低剂量CPF暴露后,能使海马的细胞生物平衡紊乱持续到青春期,并降低青春期和成年期整体情绪水平,焦虑情绪增加。Objective: The study is designed to explore the effects of prenatal animals exposed to subthreshold chlorpyrifos(CPF) on astrocyte activation in the hippocampus and neural behavioral Changes.Methods: Postnatal 11-14 days SD rats were exposed to subthreshold CPF.No acute symptoms of organophosphorus pesticide intoxicates and the animals were sacrificed at postnatal 15,20,30,and 60 days.Examining body weight gain,outward appearance of brain tissue,the coefficient of brain and the water content of brain tissue.CPF effects on hippocampal astrocytes were examined with immunohistochemical straining for glial fibrillary acidic protein(GFAP).Emotion level was scored by Emotion Level Rating Scale for Shrewmouse,Elevated plus maze was used to examine the levels of animal anxiety.Results: The outward ap-pearance of brain tissue was normal.The absolute value of body weight gain,the rate of body weight gain,the coefficient of brain and the water content of brain tissue of each experiment group were not significantly different from that of its counterpart controls(P0.05).Astrocyte activation(P0.05) in the hippocampus.The emotion level of young adult rats was significantly lower in the scores than the counterpart controls(P0.05).The anxiety level of young adult rats was increased(P0.01).Conclusion: Prenatal CPF exposure can not cause symptoms of acute organophosphate pesticide poisoning.And their body weight gain,outward appearance of brain tissue,the coefficient of brain and the water content of brain tissue are all normal.But we found that young adult animals’ emotion level was lower and high anxiety level in accompany with increased astrocyte activation in the hippocampus.
