摘  要:目的研究NK2受体mRNA在哮喘豚鼠气管、支气管及肺组织中的表达。方法用吸入卵蛋白致敏及激发诱喘方法制作哮喘豚鼠20例;正常对照组10例,以生理盐水替代卵蛋白雾化溶液吸入。用RT-PCR的方法检测肺组织中NK2受体mRNA相对含量﹔用原位杂交的方法在显微镜下观察NK2受体mRNA在气管、支气管及肺组织中表达的部位及强度同时用光密度对NK2受体mRNA相对含量进行分析。结果 NK2受体mRNA主要分布在气管黏膜下及平滑肌周围﹔哮喘豚鼠肺组织中NK2受体mRNA相对含量为(1.04±0.112),高于正常对照组豚鼠(0.796±0.30),差异有统计学意义(P<0.05);哮喘豚鼠气管、支气管黏膜下和平滑肌周围NK2受体mRNA表达的相对含量[OD(9.339±0.8177)]较正常对照组[OD(2.265±0.74)]明显增高(P<0.05)。结论哮喘豚鼠气管、支气管黏膜下平滑肌周围NK2受体mRNA相对含量表达上调,肺组织中NK2受体mRNA相对含量增多。Objective NK2 receptor is considered as one of the most important receptors of neural-mechanism of asthma.The purpose of this study was to investigate the expression of NK2 receptor mRNA in the lungs of guinea pigs with asthma.Methods Ten guinea pigs(1.5 months of age and 200~225 g weight)were sensitized by exposure to aerosolized ovalbumin and challenged by the same antigen to establish asthma model.Normal control guinea pigs(n =10)were treated with normal saline(NS)instead of aerosolized ovalbumin.The expression of NK2 receptor mRNA in the lung tissues were examined by RT-PCR.The distribution of NK2 receptor mRNA in the lungs was detected by hybridization in situ by using oligonuclectide probe.Results NK2 receptor mRNA expression could be found in the trachea,bronchus and the lung tissue in asthmatic guinea pigs.NK2 receptor mRNA was mainly located in the submucosa and around the smooth muscle of the trachea and bronchus.The expression levels of NK2 receptor in the lung and under bronchus mucosa and around smooth muscle in asthmatic guinea pigs were higher than that in the normal control(P 〈 0.05).Conclusion The expression of NK2 receptor mRNA in the lung tissues,under bronchus mucosa,and around the smooth muscle of the trachea of asthmatic guinea-pig was up-regulated.
