摘  要:为了解轮状病毒在腹泻奶牛中的流行情况,从2008年12月到2009年6月,我们在大庆某牛场共收集117份小于7日龄新生牛的腹泻标本,用A群轮状病毒抗原诊断试剂盒对腹泻标本进行检测;应用MA104细胞对阳性标本进行分离,扩增细胞分离株保护性抗原VP7基因,并对该基因进行序列分析。腹泻标本中有10份标本(8.5%)经A群轮状病毒抗原检测试剂盒确认为阳性,成功的分离出一株轮状病毒,命名为:DQ-75,分离株DQ-75VP7基因的基因型是G10,该分离株中VP7基因与现有G10型轮状病毒毒株的VP7编码区氨基酸的系统进化树分析提示其可能是随着奶牛的购入而进入我国。In order to investigate the situation of rotavirus strains circulating in diary farms,a total of 117 fecal specimens were collected from diarrhea calves under 1 week-age on a diary farm in Daqing regiona from Decmber 2008 to June 2009. Faecal specimens were tested by a Rotavirus Group A Diagnostic Kit,rotavirus positive samples was isolated by MA104 cell,VP7 sequence of the rotavirus was amplified and analyzed. The results showed that ten specimens (8.5%) were detected to be positive by a Rotavirus Group A Diagnostic Kit,one of rotavirus strains was isolated successfully by MA104 cell,genotype of the rotavirus strain was G10. Amino acid sequence and Phylogenetic analysis among the rotavirus and the existing G10 genotype rotavirus indicated that the rotavirus could be introduced into our country with the import of bovine.
