摘  要:目的:研究藻黄合剂对体外培养人肾小球系膜细胞(GMC)在高糖下分泌MMP-9、TIMP-1的影响。方法:体外培养人肾小球系膜细胞并在高糖培养基中孵育,用含有不同浓度的藻黄合剂培养液进行干预,分别观察24、48h系膜细胞MMP-9和TIMP-1的分泌。结果:藻黄合剂各剂量组在24、48h均显著升高髙糖下MMP-9的分泌(P<0.01);同时降低TIMP-1在48h的分泌(P<0.05,P<0.01)。结论:藻黄合剂可以升高高糖下系膜细胞MMP-9分泌,降低TIMP-1的分泌,这可能是藻黄合剂降解细胞外基质、防治糖尿病肾病的作用机制之一。Objective: To investigate the effect of Zaohuang Misture on expression of matrixmetalloproteinase-9 (MMP-9) and tissue inhibitor of metalloproteinases-1(TIMP-1) by human glomerular mesangial cells (GMCs) cultured under high - glucose environment. Methods: The culture of GMCs was continued under high glucose before the experiment. The changes of MMP-9 and TIMP-1 in the GMCs and in Zaohuang Misture group were examined at 24h, 48h. Results: Compared with the high glucose group, the expression of MMP-9 in Zaohuang Misture were significantly higher at 24h, 48h (P0.01); the expression of TIMP -1 in Zaohuang Misture were significantly lower at 48h (P0.05 or P0.01). Conclusion: Zaohuang Misture may modulate the process of diabetic nephropathy by changing the expression of MMP-9 and TIMP-1.
