摘 要:目的通过检测电针干预新生大鼠后大脑皮层硫化氢(H_2S)的含量,开展针刺治疗神经系统疾病的机制研究。方法 SD新生大鼠16只,日龄7 d,随机分为针刺组和对照组,每组8只。针刺组予以电针刺激百会、大椎穴,每日一次,14 d为一疗程;对照组仅固定四肢,不予针刺,共14 d。分别在针刺疗程的1、7 d和14 d对针刺组大鼠进行皮层脑血流检测。于第22天将大鼠处死,取双侧大脑皮层,应用敏感硫电极法测定大鼠脑皮层H_2S含量。结果针刺组大鼠脑皮层H_2S含量为(16.10±1.64)nmol/mg蛋白,对照组为(22.41±0.94)nmol/mg蛋白,针刺组较对照组明显降低;在疗程不同时间点,针刺均可促进皮层脑血流量增加。结论电针刺激可降低大鼠大脑局部H_2S的生成,这可能是针刺治疗神经系统疾病的气体分子学机制之一。Objective To explore the working machanism of electro-acupuncture (EA) on central nervous system by detecting hydrogen sulfide (H2S) in local cortex in neonatal rats. Methods Sixteen healthy 7 days old Sprague-Dawley neonetal rats were divided into two groups randomly (n = 8 ), EA group and control group. EA group rats were fixed and stimulated with EA at aeupoints of BAIHUI and DAZHUI for 30 minnutes daily, and kept on for 14 days. Control ones were just fixed, without aeupuncture at the same time. Regional cerebral blood flow (rCBF) was monitored before, during and after EA at different periods of intervention on dl, 7 and 14. The rats were killed at the d22. H2S in bilateral cortical tissue was measured by sensitive sulphur electrode assay. Results Compared with control group, there was an obvious reduction in H2S levels in EA group, ( 16. 10 + 1.64) nmol/mg protein vs. (22.41 ± 0. 94) nmol/mg protein, t = - 9.467, P 〈 0.01. Acupuncture increased local brain blood circulation at anytime of the stimulation. Conclusion Electro-acupuncture decreases H2S level in cortex and it might be a probable mechanism of acupuncture treatment.
