摘  要:【目的】观察黄连解毒汤对动脉粥样硬化(AS)大鼠主动脉组织中单核细胞趋化蛋白-1(MCP-1)及其特异性受体CCR2 mRNA表达的影响。【方法】选用SD大鼠,随机分为正常对照组,模型组,黄连解毒汤低、中、高剂量组(中药组,剂量分别为2.7、5.3、10.6 g.kg-1.d-1),阿托伐他汀组(剂量为1.8 mg.kg-1.d-1)。除正常对照组外,其他组均采用高脂饮食法复制AS模型,造模第6周开始给药,连续4周。取各组大鼠主动脉采用实时荧光定量PCR法检测各组MCP-1/CCR2 mRNA表达。【结果】与正常对照组比较,模型组主动脉MCP-1/CCR2 mRNA表达显著升高(P<0.05或P<0.01);与模型组比较,黄连解毒汤各剂量组MCP-1/CCR2 mRNA表达均显著降低(P<0.05或P<0.01),且呈一定的剂量效应关系,MCP-1与CCR2两者表达呈正相关。【结论】黄连解毒汤治疗AS的作用可能与其能下调MCP-1、CCR2 mRNA在主动脉的表达有关。Objective To investigate the effect of Huanglian Jiedu Decoction (HJD) on the expression of monocyte chemoattractant protein-1 (MCP-1) and its specific receptor CCR2 mRNA in rats with atherosclerosis (AS). Methods SD rats were randomized into normal control group, model group, atorvastatin ( 1.8 mg ·kg^-1·d^-1) group, and low-, moderate- and high-dosage HJD (in the dosages of 2.7, 5.3 and 10. 6 g ·kg^-1·d^-1, respectively) groups. Rats except for the normal group were given high-fat diet to induce AS. In the 6th week of modeling, the rats were given the corresponding medicine according to the experimental design for 4 continuous weeks. The expression of MCP-1/CCR2 mRNA in rats aorta was detected by the method of real-time fluorescent quantitative polymerase chain reaction (RT-PCR). Results Compared with the normal control group, the expression of aortic MCP-1/CCR2 mRNA was increased in the model group (P 〈 0. 05 or P 〈 0. 01 ). The expression of aortic MCP-1/CCR2 mRNA was significantly lower in HJD groups than that in the model group (P 〈 0. 05 or P 〈0. 01 ) , in a dose-dependent manner. The expression of MCP-1 was positively correlated with CCR2 mRNA expression. Conclusion The therapeutic mechanism of HJD for AS is probably related with the downregulation of aortic MCP-1/CCR2 mRNA.
