摘 要:应用超薄切片电镜观察,对3种番茄斑萎病毒属(Tospovirus)病毒侵染寄主细胞病理特征进行了比较分析。番茄环纹斑点病毒(Tomato zonate spot viru,s TZSV)侵染的番茄,叶部细胞中病毒粒体呈块状聚集于内质网池中,果实细胞中呈块状、管状或单个粒体散布于细胞质中,细胞核较完整,叶绿体空泡化;凤仙花坏死斑病毒(Impatiens necrotic spot virus,INSV)侵染的蝴蝶兰叶部细胞和亚细胞结构消失,病毒粒体分布较少,聚集于细胞质中的内质网池内;花生黄斑病毒(Groundnut yellow spot viru,s GYSV)侵染的辣椒果实的亚细胞结构较完整,病毒粒体呈块状聚集于细胞质的囊腔内。研究结果表明,不同种类的Tospovirus病毒侵染寄主植物后,其病毒粒体在寄主细胞内的分布、聚集方式及对亚细胞结构的影响具有明显差异,可作为诊断的依据。Using ultra-thin section electron microscopy, the cytopathology of host plants infected by three distinct Tospovirus species were studied. Tomato zonate spot virus (TZSV) aggregated in the cluster in the endoplasmic reticulum (ER) in leaf cell, but dispersed, tabulate, or single particle in the cytoplasm in fruit cells. The nuclei of the two parts are relatively integrity, and the chloroplasts are vacuolar. The leaf cell' s subcellular structures of Impatiens necrotic spot virus (INSV) -infected Phalaneaposis amabilis disappeared. INSV particles are rare under the electron microscopy, also clustered in the ER. On the contrast, the subcellular structures are relatively integrity in the fruit cell of pepper infected with Groundnut yellow spot virus (GYSV). And the massive particles clustered in the ER. These results indicated that different species of Tospovirus have considerable differences in distribution and agregation mode in the host cell, and impact on the subcellular structure, which may serve as the diagnosis criteria of Tospovirus diseases.
