摘  要:对采自河南4大山系8产地的日本三角涡虫线粒体DNA细胞色素C氧化酶Ⅰ亚基(COⅠ)基因进行PCR扩增和测序,用MEGA4.0、PAUP*4.0等程序对所得序列进行分析和系统树重建。结果表明,采自大别山系商城县的两个种群XYshangcheng1和XYshangcheng2的COⅠ片段序列同源性最高,为100%;而采自伏牛山系嵩县的种群LYsongxian与其它7种群的COⅠ片段序列同源性最低,在84.3%~89.0%之间。NJ树、MP树和贝叶斯树拓扑结构完全一致,从系统树上可以看出LYsongxian种群与其它7种群的亲缘关系最远;另外7种群又聚为2支。该聚类结果基本和各种群的山系分布相符合。同时结合基于Hsp70基因的分子系统学研究以及涡虫形态学、细胞遗传学等方面的证据初步推测:LYsongxian种群可能不是日本三角涡虫,而是三角涡虫属另外一个种,该推测是否正确尚待进一步的组织解剖学验证。In this study,COⅠ gene of 8 populations of Dugesia japonica collected from Henan Province were amplified via PCR and then sequenced.The sequences were aligned with Clustal Ⅹ 1.8,and the NJ,MP and Bayes trees were reconstructed with MEGA 4.0,PAUP4.0 and MrBayes 3.1.1,respectively.Sequences analysis shows that the homology of XYshangcheng 1 and XYshangcheng 2 was 100 %,the highest;while the homology of LYsongxian and the other 7 populations were rang from 84.3 % to 89.0 %.All the three phylogenetic trees have the same topology,indicating that the relationship of LYsongxian to other 7 populations was the remotest,it constructs one clade by itself;and the other 7 populations form the other clade with two branches being further divided.This clustering pattern is consistent with their distribution pattern in different mountains on the whole.Meanwhile,the phylogenetic tree based on another molecular marker Hsp70 gene also shows the basal position of LYsongxian population related to other Dugesia japonica populations.And combined with evidence from morphology and cell genetics,we hypothesize that this LYsongxian population might be not Dugesia japonica but another species belonging to genus Dugesia.But whether this hypothesis is true or not should be further proved by the data from histological anatomy.
