摘  要:目的检测增生性瘢痕(H)和瘢痕疙瘩(K)组织中 TGF-β_1及Ⅰ、Ⅲ型前胶原 mRNA的表达,了解其相互关系及意义。方法斑点杂交分析检测 H 和 K 组织中Ⅰ、Ⅲ前胶原及 TGF-β_1mRNA 稳态水平的改变;原位杂交检测 TGF-β_1 mRNA 在组织中的空间分布。结果①K 和 H 组织中 TGF-β_1 mRNA 稳态水平明显高于 N 组和 S 组;②K 选择性Ⅰ型前胶原 mRNA 表达增强,而 H 组织中Ⅰ、Ⅲ前胶原 mRNA 表达均增强。结论 TGF-β_1在 H 和 K 的发病机理中起了重要作用,K 和H 发生发展中具有不同的分子机理。Objective The expression ot mRNA for TGF-β_1,type Ⅰ,Ⅲ procollagen in keloid and hypertrophic scar was investigated in order to elucidate the pathogenesis of and the difference between keloid and hypertrophic scar.Method Dot-blot hybridization analysis was used to examine the type Ⅰ,Ⅲ procollagen and the expression levels of steady-state mRNA.In situ hybridization allowed direct assessment of the distribution of TGF-β_1 mRNA in the tissue.Results①Our study indicated that the expression lev- el of mRNA for TGF-β_1 significantly increased in keloid and hypertrophic scar compared with normal scar and normal skin.②In keloid tissues,type Ⅰ precollagen mRNA expression was selectively increased. However,in hypertrophic scar,type Ⅰ and Ⅲ collagen mRNAs expressions were simultaneously in- creased.Conclusion TGF-β_1 plays an important role in the pathogenesis of keloid and hypertrophic scar- ring.It implicates that distinct molecular mechanisms are operative in the development of keloid and hy- pertrophic scarring.
