摘 要:利用黄瓜绿斑驳花叶病毒(Cucumber Green Mottle Mosaic Virus,CGMMV)的特异性引物对来自广西一温室栽培的黄瓜病样进行RT-PCR检测,结果扩增得到了与预期大小相符的目的片段(650 bp)。序列分析表明,该目的片段包含有CGMMV完整的CP基因序列、部分运动蛋白基因(MP)及3'端非编码区(3'-UTR)序列,其中CP基因全长486 bp,与已报道的CP基因序列同源性为91.2%~99.4%。经系统发育分折,明确该GX-CS分离物与日本、法国、印度等分离物属于CGMMV同一类群,并推测该分离物与广西的葫芦(GX-BG)分离物具有相同的起源关系。Cucumber green mottle mosaic virus (CGMMV) was identified on cucumber (Cucumis sativus) grown in a greenhouse in Guangxi, China, by reverse transcription polymerase chain reaction (RT-PCR) with a pair of specific primers of CGMMV. A specific fragment about 650 bp in length, as was expected, was amplified by RT-PCR. Sequences analysis demonstrated that the fragment contained complete coat protein gene CP, partial movement protein gene (MP) and 3'-UTR of CGMMV. The CP gene of this Guangxi cucumber (GX-CS) isolate had 486 nucleotides in length. The CP gene showed nucleotide homology of 91.2 % to 100.0 % with other isolates or strains. Phylogenetic analyses of these sequences revealed GX-CS isolate was closely related to the Japan, France, India isolates, and maybe have same origin as Guxi GX-BG isolate.
