摘  要:目的研究何首乌提取物(PME)对百草枯引起的多巴胺神经元损伤的保护作用并探讨其机制。方法噻唑蓝(MTT)法测定细胞的存活率,尼氏染色观察小鼠多巴胺神经元,紫外吸收测定丙二醛(MDA)和超氧化歧化酶(SOD)含量,HPLC分析主要成分。结果何首乌明显改善百草枯引起的PC12细胞、小鼠多巴胺神经元的损伤、降低脑内MDA含量并增强SOD的活力。结论本研究表明何首乌提取物对百草枯引起的多巴胺神经元的损害有显著的保护作用,其作用机制可能与其抗氧化作用有关。OBJECTIVE This study investigated the protective effect of Polygonum multiflorum extract(PME) on nigrostriatal dopaminergic neuron damage caused by paraquat(PQ).METHODS MTT assayed the viability of cell.HPLC analyzed the main characteristic constituents of PME.The neurons were Nissl's stained.RESULTS PQ treatment caused a reduction of the viability of cell in vitro and nigrostriatal dopaminergic neuron lesion in the striatum.Repeated administration of PME attenuated the effect of PQ cytoxcity on dopaminergic neuron lesion and the PME character consisituents is 2,3,5,4′-tetrahydroxystilbene 2-O-D-glucopyranoside.CONCLUSION The present results suggest that PME has a beneficial effect on nigrostriatal dopaminergic degeneration induced by PQ and the mechanism associated with its antioxydation.
