摘 要:目的采用HPLC同时测定北五味子果实、藤茎超临界萃取物中五味子甲素、五味子乙素、五味子醇甲的含量。方法采用ODS-C18(4.6 mm×250 mm,5μm),以甲醇-水(75∶25)为流动相,流速:1.0 mL.min-1,检测波长为254 nm。结果五味子甲素、五味子乙素、五味子醇甲的线性范围分别为0.10~2.00μg(r=0.999 4)、0.10~2.00μg(r=0.999 7)、0.12~2.40μg(r=0.999 2),平均加样回收率分别为99.11%、101.18%、101.23%(RSD分别为1.57%、2.68%、2.80%)(n=10)。结论本方法简便准确、重现性好,能有效测定北五味子果实、藤茎超临界萃取物中五味子甲素、五味子乙素、五味子醇甲的含量。OBJECTIVE To establish a HPLC method for the determination of deoxyschizandrin,γ-schizandrin and schizandrin in supercritical carbon dioxide extract from Fructus and Caulis Schisandra chinensis.METHODS The analysis was performed on ODS-C18 column(4.6 mm×250 mm,5 μm) with the mobile phase consisting of methanol-water(75∶25) as mobile phase,flow rate of 1.0 mL·min-1.The detection wavelength was at 254 nm.RESULTS Satisfactory linearities of deoxyschizandrin,γ-schizandrin and schizandrin were shown in the range of 0.10-2.00 μg(r=0.999 4),0.1-2.00 μg(r=0.999 7)、0.12-2.40 μg(r=0.999 2).The average recovery were 99.11%,101.18% and 101.23%(RSD were 1.57%,2.68% and 2.80%)(n=10).CONCLUSION The method is simple,accurate,highly sensitive and reproducible.It can be effectly used for the determination of deoxyschizandrin,γ-schizandrin and schizandrin in supercritical carbon dioxide extract from Fructus and Caulis Schisandra chinensis.
