HPLC测定北五味子果实、藤茎超临界萃取物中木脂素的含量

摘　　要：目的采用HPLC同时测定北五味子果实、藤茎超临界萃取物中五味子甲素、五味子乙素、五味子醇甲的含量。方法采用ODS-C18（4.6 mm×250 mm,5μm）,以甲醇-水（75∶25）为流动相,流速：1.0 mL.min-1,检测波长为254 nm。结果五味子甲素、五味子乙素、五味子醇甲的线性范围分别为0.10~2.00μg（r=0.999 4）、0.10~2.00μg（r=0.999 7）、0.12~2.40μg（r=0.999 2）,平均加样回收率分别为99.11%、101.18%、101.23%（RSD分别为1.57%、2.68%、2.80%）（n=10）。结论本方法简便准确、重现性好,能有效测定北五味子果实、藤茎超临界萃取物中五味子甲素、五味子乙素、五味子醇甲的含量。OBJECTIVE To establish a HPLC method for the determination of deoxyschizandrin,γ-schizandrin and schizandrin in supercritical carbon dioxide extract from Fructus and Caulis Schisandra chinensis.METHODS The analysis was performed on ODS-C18 column（4.6 mm×250 mm,5 μm） with the mobile phase consisting of methanol-water（75∶25） as mobile phase,flow rate of 1.0 mL·min-1.The detection wavelength was at 254 nm.RESULTS Satisfactory linearities of deoxyschizandrin,γ-schizandrin and schizandrin were shown in the range of 0.10-2.00 μg（r=0.999 4）,0.1-2.00 μg（r=0.999 7）、0.12-2.40 μg（r=0.999 2）.The average recovery were 99.11%,101.18% and 101.23%（RSD were 1.57%,2.68% and 2.80%）（n=10）.CONCLUSION The method is simple,accurate,highly sensitive and reproducible.It can be effectly used for the determination of deoxyschizandrin,γ-schizandrin and schizandrin in supercritical carbon dioxide extract from Fructus and Caulis Schisandra chinensis.
