摘  要:目的:筛选成人散发性激素耐药性肾病综合征(SRNS)与激素敏感性肾病综合征(SSNS)患者间差异表达片段,获取与SRNS激素耐药相关的基因,为认识和阐明SRNS激素耐药的分子机制奠定基础。方法:应用mRNA差异显示(DDRT-PCR)技术筛选SRNS与SSNS患者外周血单个核细胞(PBMC)差异表达片段,将差异片段测序后与Genbank中人类基因序列进行同源分析。用RT-PCR验证差异片段排除假阳性。结果:获得6条SRNS与SSNS间差异片段,与Genbank中两个LOC基因、MACROD2、MAML2、NCK2和TCP1基因的部分序列高度相似。RT-PCR证实筛选的差异片段非假阳性扩增片段。结论:SRNS与SSNS间存在差异基因,该实验发现的差异基因有望成为SRNS激素耐药机制研究的新方向。Objective To investigate the differentially expressed fragments between adult steroid-resistant nephrotic syndrome (SRNS) and steroid sensitive nephrotic syndrome (SSNS).Methods The differentially expressed fragments in peripheral blood mononuclear cells (PBMC) of 6 patients with SRNS and 4 patients with SSNS were analyzed by mRNA differential display reverse transcriptase polymerase chain reaction (DDRT-PCR),and then were compared with the known homo-sapiens sequences in the Genbank.RT-PCR was used to identify the false positive.Results There were 6 differentially expressed fragments between patients with SRNS or SSNS,which were highly homologous to parts of two LOC,MACROD2,MAML2,NCK2,and TCP1 genes,and were identified by RT-PCR not to be false positive.Conclusions There were 6 differentially expressed gene fragments between SRNS and SSNS patients,which may play the role in steroid-resistant SRNS.
