摘 要:目的:建立以反相高效液相色谱法测定人血浆中米氮平浓度的方法。方法:色谱柱为KromasilC18,流动相为甲醇-水-1mol·L-1乙酸铵-5mol·L-1氨水(900:100:10:3),流速为1.0mL·min-1,内标为盐酸普萘洛尔,柱温为室温,检测波长为294nm。结果:米氮平血药浓度在20.0~500.0μg·L-1范围内线性关系良好(r=0.9990);平均回收率为98.56%,日内、日间RSD分别为3.42%~4.87%、4.25%~5.88%。结论:本方法操作简便、回收率高、精密度好,可用于米氮平的治疗药物监测。OBJECTIVE:To develop an RP-HPLC method for the quantitative determination of mirtazapine in human plasma.METHODS:The separation was performed on Kromasil C 18 (150 mm×4.6 mm,5 μm)column with the mobile phase consisted of methanol-water-1 mol · L-1 ammonium acetate-5 mol · L-1 ammonium hydroxide(900:100:10:3)using propranolol hydrochloride as internal standard.The flow rate was 1.0 mL · min-1 ;the detection wavelength was 294 nm and column temperature was room temperature.RESULTS:Mirtazapine had a good linear relationship in the range of 20.0~500.0 μg · L-1 (r=0.999 0)with an average recovery of 98.56%.The RSD of intra-day and inter-day were 3.42%~4.87% and 4.25%~5.88%.CONCLUSION:The method is simple,and of high recovery and good precision for therapeutic drug monitoring of mirtazapine.
