Research On Tissue Culture of Peach Cultivars——A case study of "Wujingbaifeng" Peach and "Chuanzhongbai" Peach

摘  要:选取从日本引进的"武井白凤"和"川中岛白桃"2个桃品种为试验材料,进行了桃茎尖和茎段培养的研究,探讨了取材时期、取材部位、消毒方法、激素配比、大量元素和活性炭对桃组织培养的影响。研究结果表明:"武井白凤"最适诱导培养基为MS+6-BA1.5mg·L-1+IAA0.2mg.L-1,"川中岛白桃"最适诱导培养基为MS+6-BA1.5mg·L-1+IAA0.1mg·L-1,且取4月、5月的茎段用2%NaClO消毒15min时建立桃外植体最好。桃茎段初代培养时在培养基中加入1%的活性炭对桃组织培养的诱导有益,添加2%的活性炭时则会抑制出芽。将桃茎段外植体接种在MS培养基上时,出芽数和出芽率均高于接种在1/3、2/3MS(大量元素)培养基上。Taking two peach cultivars"Wujingbaifeng" and imental materials, this paper makes research on culture of "Chuanzhongbai" imported from Japan as exper peach shoot tips and stems,and discusses the in fluence of selecting periods, selecting parts, sterilizing ways,hormone combination, macro-element and activated carbon on peach tissue culture. The results are as follows: (1)The optimum medium of "Wuiingbaifeng"peach shoot propagation during the primary culture is MS+ BA1. 5mg ·L^-1 +IAA0. 2mg ·L^-1 and that of"Chuanzhongdao" is MS+BA1. Smg ·L^-1+IAA0.1mg ·L^-1,and the best peach explants are proved to be the stems selected in April or May and sterilized by NaCIO(2%)for fifteen minutes. (2)It is beneficial for the induction of stems to add 1 % activated carbon to the culture medium during the primary culture,whereas adding 2%activated carbon can inhibit the budding growth. (3)The budding number and rate of MS medium are higher than that of 1/2 and 2/3 MS medium.
