不同剂量左旋精氨酸对骨质疏松大鼠血生化的影响

摘  要:目的探讨不同剂量左旋精氨酸(L-FArg)对骨质疏松大鼠血生化的影响。方法将50只3月龄雌性大鼠随机分为假手术组(Sham组)10只和卵巢切除术组(OVX)40只,对OVX组进行去势造模,12周后随机抽取10只进行骨密度测量,证实骨质疏松模型建立成功后,再随机分为4组(骨质疏松组,低剂量组,中剂量组,高剂量组),分别以不同剂量的L-Arg皮下注射。12周后下腔静脉取血测量血清Ca,P,AKP,OC等指标。结果 (1)大鼠去势12周后,OVX组BMD较Sham组降低(P<0.01),确定骨质疏松模型建立成功。(2)给药12周后,血清ALP值:OP组ALP较Sham组升高(P<0.05);ML-Arg组、HL-Arg组ALP较OP组降低(P<0.05);LL-Arg组与OP组相比无差异(P>0.05)。血清OCN值:OP组OCN较Sham组升高(P<0.05);LL-Arg组、HL-Arg组OCN较OP组升高(P<0.05);ML-Arg组与OP组相比无差异(P>0.05)。低剂量组和中剂量均呈现低钙高磷表现,高剂量组呈现低磷的表现。结论不同剂量的L-Arg对骨质疏松大鼠血生化产生不同的影响,可以根据血生化值调整L-Arg使用量,使用L-Arg防治绝经后骨质疏松同时注意补钙。Objective To explore the effects of different doses of L-Arginine to osteoporosis rats blood biochemical.Methods 50 cases of 3 month old female rats were randomly divided into two group which were sham-operated group(n=10)and ovarietomy group(OVX)(n=40).Making model to OVX using the method of resection ovaries,after 12 weeks randomly selected 10 to measure of bone mineral density(BMD),to confirm the osteoporosis model was set up successfully,then were randomly divided into 4 group(osteoporosis-group,low dose group,medium dose group,and high-dose-group),hypodermic injection with different doses of L-Arg.After 12 weeks,to measure the value of serum Ca,P,AKP,OC from the inferior vena cava blood.Results After the rats were castrated for 12 weeks,the BMD of the OVX group drops compared to the Sham group(P0.01).Make sure the osteoporosis model of Castrated rats was successfully established.After 12 weeks the injection,compared to the Sham group,the serum ALP of the OP group rose(P0.05),the value of the ML-Arg group and HL-Arg group droped compared to the OP group(P0.05).And the ALP value of the LL-Arg group is of no significant difference with the OP group.Compared to the Sham group,the blood OCN of the OP group rose(P0.05),and the value of LL-Arg group and the HL-Arg group rose compared to the OP group(P0.05).There was no significant difference between the ML-Arg group and the OP group(P0.05).The blood biochemical value displayed low calcium and high phosphorus in low dose group and middle-dose-group,and low-phosphorus in high-dose group.Conclusion The blood biochemical value is different through feeding osteoporosis rats with different doses of L-Arg.The dose of L-Arg were adjusted according to blood biochemical values.When L-Arg was used the calcium should be complement to prevent Postmenopausal osteoporosis.
