摘 要:目的建立一种测定人血浆中谷氨酸(glutamic acid,Glu)浓度的高效液相色谱法(high-performance liquid chromatog-raphy,HPLC)。方法血浆经高氯酸沉淀蛋白,6-氨基喹啉-N-羟基琥珀酰亚氨基甲酸酯(AQC)衍生其中的氨基酸后进行HPLC分离测定。结果在浓度5~320μM范围内谷氨酸线性关系良好(r>0.999),其日内、日间精密度(RSD)均低于10%,低、中、高浓度质控样品的加样回收率均超过95%。用该法测定了20名健康志愿者血浆谷氨酸浓度,平均为(51.74±16.45)μM。结论本实验所建立的HPLC法灵敏、快速,结果准确可靠,适用于临床大样本和中枢神经疾病研究中谷氨酸的测定。Objective A sensitive,reproducible,and robust high-performance liquid chromatography(HPLC) method has been validated for determining concentration of glutamic acid in human plasma.Methods Proteins in plasma were precipitated with perchloric acid,followed by derivatization with 6-aminoquinolyl-N-hydroxysuccinimidyl carbamate(AQC).Separation of glutamic acid was achieved using reversed-phase HPLC conditions and ultraviolet detection.Results Excellent linearity was observed for glutamic acid over its concentration ranges with correlation coefficients(r〉0.999).The intra-and inter-day precision were within±10%.This method utilizes quality controls and demonstrates excellent plasma recovery and improved accuracy.The developed method has been successfully applied to measure plasma glutamic acid in twenty volunteers(51.74±16.45)μM.Conclusion This assay is well suited for quantitative determination of glutamic acid in clinical studies,and also can be used to support investigations of some central nervous disease in patients.
