摘 要:目的比较甘油和氯化汞所诱发的急性肾衰模型对家兔酚红药动学的影响,为选择较佳的肾小管衰竭模型提供依据。方法家兔随意分成正常组,以及甘油和氯化汞诱发的两个急性肾衰模型组。静脉注射1.2 mg.kg-1酚红后,测定血药浓度,计算酚红药动学参数。结果与正常组比较,甘油和氯化汞诱发的肾衰模型组的血清尿素氮(blood urea nitrogen,BUN)和肌酐(creatinine,Cr)显著性增高(P<0.01),氯化汞组的BUN和Cr较甘油组增高约3.8倍(P<0.01),各组之间的c(BUN):c(Cr)无显著性差异。与正常组比较,2个模型组的消除速率常数(kel)显著性下降(P<0.01),消除半衰期(t1/2)显著性延长(P<0.01),以氯化汞组的变化更显著。模型组的酚红总清除率(CLt)较正常组显著性减少(P<0.01)。氯化汞组的表观分布容积(Vd)较甘油组和正常组显著性增大(P<0.01)。结论 2种模型组均可导致肾小管排泌功能下降,氯化汞所致的急性肾衰更显著。Objective To investigate the pharmacokinetics of phenolsulphonphthalein(PHP) on the glycerol and mercuric chloride induced acute renal failure(ARF) rabbits in order to search a more suitable model for inducing renal tubular failure.Methods Rabbits were divided into three groups randomly,a control and the glycerol and mercuric chloride induced ARFs,which were administered with 1.2 mg·kg-1 PHP intravenously,and the plasma concentrations were detected for calculating the pharmacokinetic parameters.Results Compared with control group,the blood urea nitrogen(BUN) and creatinine(Cr) concentrations were increased significantly in both ARFs(P0.01).The BUN and Cr values in mercuric chloride group was 3.8-fold higher than that of the glycerol(P0.01),and c(BUN):c(Cr) value was not altered significantly in each group.The elimination rate constant(Kel) of PHP in the two ARF groups were significantly lower than the control(P0.01),and the elimination half-life(t1/2) were significantly longer(P0.01),in which the mercuric chloride group altered more than the glycerol(P0.01).The total body clearance(CLt) of PHP in ARFs decreased significantly compared to the control(P0.01).The apparent volume of distribution(Vd) of PHP in mercuric chloride group enlarged more than the glycerol and the control(P0.01).Conclusions The renal tubular secretion is impeded heavily in both ARF models,and the mercuric chloride is more severe than the glycerol.
