摘 要:根据细菌磁颗粒对植物病毒颗粒的非特异性吸附作用,建立了一种新的提取植物病毒RNA的方法。利用细菌磁颗粒收集黄瓜绿斑驳花叶病毒、黄瓜花叶病毒和百合无症病毒等3种病毒颗粒,富集分离后,通过高温使其裂解释放出病毒RNA,进行RT-PCR检测。与利用Trizol试剂提取这3种植物病毒的RNA相比,具有操作简单、无毒性,能够显著缩短提取时间。对黄瓜绿斑驳花叶病毒的检测灵敏度验证表明:BMPs-RT-PCR与Trizol-RT-PCR的检测灵敏度相同,均为105倍的植物叶片组织稀释液。A novel method used to isolate RNA of the plant virus was established,which was based on the non-specific absorption between the bacterial magnetic particles(BMPs)and the plant viruses.The viriones of Cucumber green mottle mosaic viruses(CGMMV),Cucumber mosaic virus(CMV),and Lily symptomless virus(LSV)were enriched by bacterial magnetic particles,respectively,and RNAs were released by a applied high temperature.RT-PCR method was used for detection CGMMV,CMV,and LSV.Compared to the extraction method of RNA by utilizing the organic reagents(Trizol reagent),the BMPs-based method for RNA collection has the advantages of simplicity,nontoxicity and time saving.The sensitivity comparison of the CGMMV detection indicates that BMPs-based RT-PCR and Trizol-based RT-PCR had the same sensitivity of the 105 dilution times of the plant leaf materials.
