摘　　要：目的探讨胰岛素样生长因子（ＩＧＦｓ）多肽、受体和结合蛋白在免疫调节和免疫发育中的意义。方法采用逆转录聚合酶链反应技术在ｍＲＮＡ水平观察正常成人和新生儿淋巴细胞静息和ＰＨＡ刺激不同时间ＩＧＦｓ及其受体，结合蛋白基因的表达。结果在静息和活化的成人和新生儿淋巴细胞均有ＩＧＦ－Ⅰ、ＩＧＦ－ⅠＲ、ＩＧＦ－ⅡＲ、ＩＧＦＢＰ－２、３、４、６基因表达，并随ＰＨＡ刺激时间的延长表达水平有所变化；ＩＧＦＢＰ－５ｍＲＮＡ在活化的淋巴细胞表达；ＩＧＦ－Ⅱ、ＩＧＦＢＰ－ⅠｍＲＮＡ仅在新生儿淋巴细胞检测到。结论ＩＧＦｓ可能通过参与淋巴细胞的增殖、分化，影响免疫系统的功能和免疫发育过程。Purpose Gene expression of insulin like growth factors (IGFs) in human peripheral and cord blood lymphocytes were observed to study its significance. Methods Using reverse transcriptase polymerase chain rea ction technique,we observed the expression of IGFs, IGF receptors and IGF binding proteins in the rest and PHA stimulated lymphocytes at mRNA level. Results IGFⅠ,IGFⅠR,IGFⅡR,IGFBP2,3, 4 and 6 mRNA were found in both rest and activated lymphocytes of human peripheral and cord blood.Compared with the rest lymphocytes, the PHA stimulated lymphocytes had lower level of IGFBP-3 and 4 mRNA and higher mRNA level of IGFⅠ,IGFⅠR,IGF ⅡR,IGFBP2, and 6.Moreover,with the stimulation time of PHA,the expression of IGFBP2 was increasing,while IGFBP3 was decreasing.IGFBP5 was only detected in the activated lymphocytes.Both IGFⅡ and IGFBPⅠ could only be detected in the rest and activated cord blood lymphocytes. Conclusions IGFs may be involved in the process of proliferation and differentiation of lymphocytes and the modulation of the immune functions and immune development.
