摘  要:目的:阐明氯化甲基汞(MMC)致脑发育损伤与海马组织蛋白激酶C(PKC)活性变化之间的关系,探讨MMC致脑发育损伤机制。方法:选用Wistar雌性大鼠120只,随机分为3个MMC剂量实验组和1个对照组,每组30只,实验组母鼠从妊娠前90d至仔鼠出生后30d连续喂饲含有不同剂量MMC(0.75,1.50和3.00mg·kg-1)的普通饲料,取其生后(PND)3、7、14、21和30d仔鼠海马组织提取胞浆和胞膜PKC。采用改良Takai法观察MMC对发育大鼠海马PKC活性的影响。结果:随着脑汞含量的增加,仔鼠实验组和对照组海马组织胞膜胞浆PKC的活性随仔鼠生后时间的延长,逐渐增高,生后30d达峰值。1.50mg·kg-1剂量组生后21和30d仔鼠以及3.00mg·kg-1剂量组仔鼠海马组织胞膜和胞浆PKC活性较对照组明显增高(P<0.05)。结论:长期接触低剂量MMC可引起发育阶段大鼠海马组织胞浆及胞膜PKC活性升高。MMC可能通过影响大鼠脑发育过程中PKC活性介导其神经毒性作用。Objective To study the effects of methylmercury chloride(MMC)on protein kinase C(PKC)activity in rat developing hippocampus.Methods The animals in experimental groups were fed with standard rat chow including 0.75mg·kg-1 MMC,1.5mg·kg-1 MMC and 3.0mg·kg-1 MMC respectively for 90dbefore gestation to 30dpost parturition.Hippocampus of pups from each group on postnatal days(PND)3,7,17,21 and 30were dissected.All the samples were separated into cytosol and membrane subcellular fractions and assayed for PKC activity by the improved Takai's method.Results The membrane and cytosolic PKC activities of pup hippocampus in experimental groups were significantly higher than those in control group.The PKC activities of rats in 3.0mg·kg-1 MMC group and PND 21,30in 1.5mg·kg-1 MMC group were significantly increased compared with control group(P〈0.05).Conclusion The damage of the rat developing hippocampus by chronic exposure to MMC is possibly correlative with the enhanced effect of MMC on PKC activity.
