摘  要:目的探讨厄贝沙坦对肾性高血压大鼠(RHR)肾脏腺苷三磷酸酶活性及血管紧张素Ⅱ(AngⅡ)的影响。方法采用两肾一夹制造RHR模型18只随机分为模型组、厄贝沙坦组、停药组,每组6只,未造模的6只大鼠为假手术组,用药8周后,用酶学比色法检测大鼠左、右肾皮质Na^+-K^+-ATP酶、Ca^(2+)-ATP酶活性,放射免疫技术测AngⅡ水平。结果与假手术组比较,模型组、厄贝沙坦组和停药组大鼠左右肾皮质Na^+-K^+-ATP酶活性显著降低,血浆AngⅡ显著升高,模型组和停药组Ca^(2+)-ATP酶活性显著降低;与模型组比较,厄贝沙坦组Ca^(2+)-ATP酶活性显著升高,厄贝沙坦组和停药组血浆AngⅡ显著升高;与厄贝沙坦组比较,停药组Ca^(2+)-ATP酶活性显著降低,右肾皮质AngⅡ显著升高。肾脏Ca^(2+)-ATP酶与局部AngⅡ呈显著负相关。结论厄贝沙坦影响RHR肾脏Ca^(2+)-ATP酶活性及AngⅡ水平,停药后AngⅡ及Ca^(2+)-ATP酶活性变化,可能是高血压反跳的机制之一。Objective To explore the effects of irbesartan on activities of Na^+-K^+-ATPase,Ca^2+- ATPase and level of Ang Ⅱ in kidney of RHR. Methods Renovascular hypertension was induced by two kidney-one clip method. Eighteen RHR were randomly divided into RHR group(Mod), irbesartan group(Irb) and discontinuation of medication group(Dis). Other 6 SD rats served as sham operation group(Sha). ATPase activity and Ang Ⅱ content in renal cortexes of RHR were determined by spectrophotometry and radioimmunoassay respectively after 8 weeks of irbesartan treatment. Results Compared with Sha,the activities of Na^+-K^+- ATPase of bilateral renal corte- xes were significantly lower, plasma Ang Ⅱ level was significantly higher in Mod, Irb and Dis, the activity of Ca^2+-ATpase was significantly lower in Mod and Dis. Compared with Mod, the activity of Ca^2+-ATPase significantly increased in Irb,plasma Ang Ⅱ significantly higher in Irb and Dis. Compared with Irb, the activity of Ca^2+-ATPase decreased and Ang Ⅱ level significantly increased in Dis. There was a significant negative correlation between Ang II and the activity of Ca^2+- ATPase in kidney. Conclusions Irbesartan influences the activity of Ca^2+-ATPase and level of Ang Ⅱ in kidney of RI-IR. The Ang Ⅱ and activity of Ca^2+-ATPase are in disorder after discontinuation of medication, which might be one of the mechanisms of rebound of hypertension.
