摘  要:目的:建立金银花叶中绿原酸的含量测定方法,考察不同型号的大孔吸附树脂对金银花叶中绿原酸的提取纯化效果,为金银花药材的综合开发利用提供参考。方法:采用高效液相色谱法测定指标成分。采用静态吸附法和动态吸附法,考察大孔树脂的吸附、洗脱附性能和纯化效果。结果:绿原酸在2.376 8~60.476 9μg与峰面积呈良好线性关系,平均加样回收率96.82%,RSD 1.83%。HPD450大孔吸附树脂纯化效果较好,优化条件为:柱径比3∶1,药液质量浓度0.5~1.0 g·mL-1、吸附速度0.5 BV·h-1、树脂吸附量生药0.3 g·g-1、洗脱溶媒30%乙醇、洗脱速度2 B V·h-1,洗脱溶媒用量2 BV。结论:HPD450大孔树脂对绿原酸纯化效果较好,工艺稳定可行,可用于工业化生产。Objective:To establish a method for determination of chlorogenic acid in Lonicera japonica leaves by HPLC,and study on extracting and purifying chlorogenic acid from Lonicera japonica leaves by different macroporous resin,so as to provide a reference for the comprehensive development and utilization of honeysuckle herbs.Method: The chlorogenic acid was determined with the detection wavelength 327 nm by HPLC.Static adsorption and dynamic adsorption were used to optimize the adsorption and elution process by macroporous resin.Result:Chlorogenic acid was linear in the range of 2.376 8-60.476 9 μg.The average recovery was 96.82%,RSD was 1.83%.At the same time,the purification effect of chlorogenic acid by HPD450 macroporous resin was better than other resins,the optimal conditions were:column diameter ratio 3 ∶ 1,liquid concentration 0.5-1.0 g·mL-1,absorption velocity 0.5 BV·h-1,resin adsorption quantity 0.3 g·g-1,elution solvent 30% ethanol,elution speed 2 BV·h-1,elution solvent volume 2 BV.Conclusion:The process of purification of chlorogenic acid from Lonicera japonica leaves by HPD450 macroporous resin was stable and could be used for industrial production.
