摘  要:目的调查多药耐药铜绿假单胞菌中接合性质粒、转座子、整合子等可移动遗传元件的存在情况。方法收集医院分离自痰液和伤口分泌物标本的铜绿假单胞菌共20株,采用聚合酶链反应(PCR)及序列分析的方法分析接合性质粒遗传标记(traA、traF、trbC)、转座子遗传标记(tnsA/Tn7、tnpA/Tn21、ISCR1、ISCR3/14、IS1133、IS26、ISpa7、ISkpa6、ISkpn7)、整合子遗传标记(intⅠ1、intⅠ2、intⅠ3),共15种可移动遗传元件。结果 20株铜绿假单胞菌共检出tnpA/Tn21、IS26、ISpa7、intⅠ1等4种基因,阳性株数分别为11株(55.0%)、2株(10.0%)、7株(35.0%)、11株(55.0%),其余11种基因未检出;只有8株(40.0%)未检出所进行检测的15种可移动遗传元件,但12株(60.0%)携带了>1种可移动遗传元件;阳性检出基因可分为6种构成模式,其中(tnpA/Tn21+ISpa7+intⅠ1)模式检出率最高,为6株(30.0%)。结论 20株铜绿假单胞菌未检出接合性质粒遗传标记,但检出的3种转座子和1种整合子遗传标记可能与铜绿假单胞菌呈多耐药的表型相关。OBJECTIVE To investigate the distribution of mobile genetic elements: plasmids, transposons, and integrons in multidrug-resistant Pseudornonas aeruginosa (P. aeruginosa) strains. METHODS A total of 20 strains of muhidrug-resistant P. aeruginosa isolated from sputum and wound secretion were collected, Then, 15 kinds of mobile genetic elements (traA, traF, trbC, tnsA /Tn7, tnpA/Tn21, ISCR1, ISCR3/14, ISl133, IS26, ISpaT,ISkpa6, ISkpnT, int I 1, int I 2, int I 3) were analyzed by PCR and verificated by DNA sequencing. RESULTS In 20 strains of P. aeruginosa, 11 strains(55.0%) ,2 strains(10.0%) ,7 strains(35.0%)and 11 strains (55.0%)were detected to carry tnpA/Tn21, IS26, ISpaT, int I 1, respectively. However, the rest 11 kinds of genes couldn't be detected out. Moreover, 8 strains(40. 0%) were not carring mobile genetic elements, but 12 strains(60.0%) were detected to carry at the least a kind of mobile genetic element. Furthermore, positive genes could be divided into 6 kinds of positive modes, among which the detection rates of the mode (tnpA/Tn21+ISpa7 +intI 1) were the highest with 6 strains(30. 0%). CONCLUSION Plasmids are not detected in 20 strains of multidrug-resistant P. aeruginosa, but S kinds of transposons and 1 kind of integron were detected,which might play a role in muhidrug resistant phenotype.
