摘 要:观察大鼠肠缺血-再灌流过程中小肠、肝、肺组织TNFα和IL-6mRNA表达的规律,发现肠缺血1h,小肠TNFα和IL-6mRNA表达开始增加,再灌流后0.5~1h表达至峰值,并且与组织及血浆中蛋白水平变化趋势一致;与小肠相比,肝、肺组织TNFα和IL-6mRNA表达升高的时相晚,且与血浆TNFα浓度变化不一致。结果表明,小肠是肠缺血-再灌流后较早产生TNFα和IL-6的部位。提示小肠在肠缺血-再灌流后在全身性炎症反应综合征的启动中可能起重要作用;血中TNFα浓度不能完全反映不同组织TNFα的含量,组织局部TNFα的增加可能是介导组织损伤的机制之一。The expression regularity of TNFα and IL 6 mRNA in small intestine, liver and lungs after ischemia reperfusion in rats was observed. It was revealed that the expression of TNFα and IL 6 mRNA in small intestine was enhanced at 1h post ischemia, peaking at 0.5~1h after reperfusion, which was parallel to the changes in plasma and tissue TNFα and IL 6 levels. Compared to small intestine, the enhanced expression of TNFα and IL 6 mRNA was delayed and lower in liver and lungs. Changes in pulmonary TNFα content were not in accordance with that of plasma TNFα level. These data indicated that small intestine might serve as a cytokine generating organ during the early stage after local ischemia reperfusion, thus playing an important role in promoting SIRS; the systemic TNFα levels did not reflect the local contents in various tissues, and the elevation of TNFα in tissues might be one of the important mechanisms inducing organ dysfunction.
