摘 要:目的探讨脂联素对动脉钙化的作用。方法体外培养脂联素基因敲除(Adiponectin-/-)小鼠主动脉平滑肌细胞(ASMCs),加脂联素干预,采用α-磷酸奈酚法测定碱性磷酸酶(ALP)活性,放射免疫法检测骨钙素含量,Westernblot检测Runx2表达变化,1%茜素红进行钙化结节染色。结果脂联素通过抑制ALP活性、骨钙素分泌、下调Runx2表达,从而抑制细胞钙化结节形成。结论脂联素抑制ASMCs向成骨细胞表型分化,保护动脉钙化。【Objective】To investigate the effects of adiponectin on arteriosteogenesis in vitro.【Methods】The ASMCs of Adiponectin-/-mice in culture were treated with adiponectin.ALP activity was determined by using pnitrophenyl phosphate as substrate.Osteocalcin was measured by radioimmunoassay.Runx2 protein expression was analyzed by Western blot.The calcified nodules were stained by Alizarin Red S.【Results】Adiponectin suppressed ALP activity,osteocalcin secretion,Runx2 protein expression,and the formation of mineralized nodules in cultured ASMCs.【Conclusion】Adiponectin inhibits ASMCs calcification in vitro and may play a protective role against arterial calcification.
