摘  要:目的:探讨阿托伐他汀预处理对大鼠心肌缺血再灌注后的作用及其可能的机制。方法:42只Wistar大鼠随机分为3组:假手术组、缺血再灌注组、阿托伐他汀预处理组;建立大鼠心肌缺血再灌注损伤模型,测定肌酸激酶(CK)水平,Evans b lue、TTC染色后,用计算机图像分析软件计算梗死面积;测定心肌髓过氧化物酶(MPO)、血清白介素-6(IL-6)及白介素-8(IL-8)含量。结果:与缺血再灌注组比较,阿托伐他汀预处理组CK减少,心肌梗死面积显著减小;M PO、IL-6、IL-8显著降低。结论:阿托伐他汀能明显减轻心肌再灌注损伤的发生和发展,其作用机制可能通过抑制炎症因子的合成,减少中性粒细胞的浸润,从而改善心肌缺血再灌注损伤的发生。Objective: To investigate the protective effects of atorvastatin and its mechanisms in the treatment of myocardial ischemia reperfusion injury in rats.Methods: forty-two Wistar rats were randomly divided into three groups,sham group,ischemia reperfusion injury group and atorvastatin preconditioning group,and then treated accordingly.The MIRI model was produced;the activities of interleukin-6,interleukin-8 creatine kinase,myeloperoxidase were detected.After reperfusion,the area at risk was determined using Evans blue and infarct area using triphenyl tetrazolium chloride staining respectively.Results: in comparison with MIRI group,atorvastain decreased IL-6、IL-8、CKand MPO.Infarct size were reduced.Conclusion: atorvastatin appears to have an acute protective effect in a rat model of ischemia-reperfusion.The mechanism of protection is reduced systemic inflammatory responses.
