摘 要:应用新型调制叶绿素荧光成像系统Imaging-PAM,通过最大叶绿素荧光量子产量(Fv/Fm)、实际荧光量子产量(YⅡ)指标的测定原位评估了友谊河底泥对斜生栅藻(Scenedesmus obliqnus)的毒性效应.数据显示,在友谊河底泥悬浮液中,栅藻的Fv/Fm和YⅡ值在12h前快速上升,12h后呈现出时间-效应型曲线,72h时Fv/Fm和YⅡ分别下降至对照的44%和35%,Fv/Fm和YⅡ的半抑制时间分别为57h和45h.同时,通过对友谊河底泥样品中重金属含量的检测,计算地积累指数(Igeo),得出友谊河底泥中的Hg、Zn和Cu分别为严重污染,重污染和偏重污染水平,这可能是影响藻类光合作用的主要污染物.本研究为进行底泥毒性评估提供了新的原位分析方法。New modulated chlorophyll fluorescence imaging system Imaging-PAM has been applied as an effective detection in short-term bioassays with Scenedesmus obliquus.Maximum quantum yield of chlorophyll fluorescence(Fv/Fm) and the actual fluorescence quantum yield(YⅡ) were used to assess the toxicity of the sediment from Youyi River.Data showed that in 12h exposure there was a rapid increase in Fv/Fm and YⅡof Scenedesmus obliquus in the sediment suspension of Youyi River,whereas after 12h a time-response curve appeared.In 72h the Fv/Fm and YⅡdropped to 44% and 35% of that of the control respectively,the halfinhibition time of Fv/Fm and YⅡwere 57h and 45h.At the same time by detecting the concentration of heavy metals in the sediment of Youyi River,the index of geoaccumulation(Igeo) was accounted as follows:Hg,Zn,and Cu reached serious pollution,heavy pollution and partial heavy pollution levels respectively.They are the probobly the primary contaminants affecting the photosynthesis of algae.This study may provide a new method to assess the toxicity of sediment in situ.
