RP-HPLC法同时测定四神丸中补骨脂素、异补骨脂素、五味子醇甲和吴茱萸碱的含量

摘　　要：目的:建立以反相高效液相色谱法同时测定四神丸中补骨脂素、异补骨脂素、五味子醇甲及吴茱萸碱含量的方法。方法:色谱柱为Diamonsil C18(200mm×4.6mm,5μm),流动相为甲醇-乙腈(93∶7)和0.1%磷酸水溶液(梯度洗脱),检测波长为225nm,柱温为25℃。结果:补骨脂素、异补骨脂素、五味子醇甲、吴茱萸碱的进样量分别在1.560×10-4～1.560×10-3、1.440×10-4～1.440×10-3、5.590×10-5～5.590×10-4、2.080×10-5～2.080×10-4mg范围内与各自峰面积积分值呈良好线性关系(r均>0.9990);四者平均回收率分别为101.7%(RSD=2.0%,n=9)、99.0%(RSD=2.0%,n=9)、100.0%(RSD=2.0%,n=9)、97.3%(RSD=1.4%,n=9)。结论:本法操作简便、结果准确,可用于同时测定四神丸中4种成分的含量。OBJECTIVE：To establish RP-HPLC method for the content determination of psoralen,isopsoralen,schisandrin and evodiamine in Sishen pills.METHODS：The determination was performed on Diamonsil C 18（200 mm×4.6 mm,5μm）column.The mobile phase consisted of methanol-acetonitrile（93：7）and 0.1% phosphoric acid solution（gradient elution）.The detection wavelength was set at 225 nm and column temperature was 25℃.RESULTS：The linear range was 1.560×10 -4 ～1.560×10 -3 mg for psoralen,1.440×10 -4 ～1.440×10 -3 mg for isopsoralen,5.590×10 -5 ～5.590×10 -4 mg for schisandrin,2.080×10 -5 ～2.080×10 -4 mg for evodiamine（r0.999 0）.The average recoveries were 101.7%for psoralen（RSD=2.0%,n=9）and 99.0%for isopsoralen（RSD= 2.0%,n=9）and 100.0% for schisandrin（RSD=2.0%,n=9）and 97.3%for evodiamine（RSD=1.4%,n=9）.CONCLUSION：The method is simple and accurate for the content determination of 4 components of Sishen pills.
