摘  要:目的:探讨复方中药抗肿瘤的机制及作用靶点。方法:造模后,荷瘤鼠随机分成6组,分别给生理盐水(空白组)、养阴清热方——安体优Ⅰ号低(ATU-L)、中(ATU-M)、高(ATU-H)3个剂量组灌胃、及阳性对照α-IFN和紫杉醇(PTX)组治疗,第14、21天处死动物,制备相应标本,以聚丙烯酰胺凝胶电泳—银染法及TRAP-PCR-ELISA法,定性、定量测定端粒酶活性。结果ATU-H组及ATU-M组对荷瘤小鼠的端粒酶活性有一定抑制作用,在治疗第14天及第21天,ATU-H组端粒酶活性均为阴性,ATU-L组均为阳性,而ATU-M组在第14天为阳性,在第21天为阴性,电泳银染法也显示了相近结果。结论:安体优Ⅰ号对615小鼠HCa-F肝癌的端粒酶活性具有一定的抑制作用。Objective:To discover the anti-tumor mechanism and action target of Traditional Chinese Medicine ATU-Ⅰ on the basis of observing its inhibition of subcutaneous transplanting liver carcinoma(HCa-F) of recombinant in-bred strain 615 mice.Methods:All the tested carcinoma model mice are grouped into 6 groups which were administered with N.S.(model group/negative control),AUT-I low dose(ATU-L),AUT-I medium dose(ATU-M),AUT-I high dose(ATU-H),(-IFN(positive control 1),and Pacilitaxel(positive control 2) respectively.Keep observing the change of tumor volume and survival time until the 14thand 21st day when some of the tested mice were killed,tumors were weighed,and the relative sample were made respectively.The activity of telomerase was measured qualitatively and quantitatively through PAGE-silver-staining assay and TRAP-PCR-ELISA assay.Results :The activity of telomerase was inhibited in ATU-H group and ATU-M group.On day 14 and day 21 of administration,research has showed that the telomerase activity was negative in ATU-H group and positive in ATU-L group.Whereas ATU-M group,it showed positive on day 14 and negative on day 21.Similar result was shown with PAGE-silver-staining assay.Conclusions: ATU-I can partly inhibit the activity of telomerase of 615 mice with HCa-F liver carcinoma.
