摘  要:目的:从白首乌块根中提取纯化C21甾体苷类化合物,建立相关检测方法,并研究其抗肿瘤活性。方法:运用高效离心分配色谱,从白首乌总苷中分离纯化C21甾体苷,提取物通过HPLC分析,并利用MTT法进行细胞毒活性测试。结果:在PC-3,SGC-7901,BGC-823,BEL-7402和KB细胞株上进行的测试表明,提取物对PC-3和SGC-7901具有抗肿瘤作用,IC50分别为(29.58±1.65)μg/mL和(66.55±2.73)μg/mL。结论:运用高效离心分配色谱分离纯化白首乌中的C21甾体苷类化合物,操作简单,成分明确,提取物表现出一定抗肿瘤活性。Objective:To extract and purify of C21 steroidal glycosides from the root of Cynanchum auriculatum,es-tablish a related method for detection and study their antineoplastic activities.Methods:By using high performance cen-trifugal partition chromatography,isolate and purify of C21 steroidal glycosides from the crude steroides of Cynanchum auriculatum.The extract was detected with HPLC and tested by using MTT method to study the cytotoxic activities.Results:In testing cytotoxicity against human cancer cell lines PC-3,SGC-7901,BGC-823,BEL-7402 and KB,the extract showed inhibition against two tumor cell lines:PC-3 and SGC-7901,with IC50 being(29.58 ± 1.65) μg/mL,(66.55 ±2.73) μg/mL respectively.Conclusion:The method was proved to be simple,accurate and composition-defi-nite by HPCPC,and the extract showed moderate antineoplastic activity against human cancer cell lines.
