芪蛭皱肺无糖颗粒对慢性阻塞性肺疾病大鼠模型小气道重塑的影响

摘　　要：目的:通过观察芪蛭皱肺无糖颗粒(黄芪、水蛭、党参、丹参、麦冬等)对患慢性阻塞性肺疾病(COPD)大鼠细胞外基质中层黏连蛋白(LN)、透明质酸(HA)水平的变化和小气道病理改变来探讨小气道重塑的影响。方法:两次气管内注入脂多糖及每日熏香烟的复合刺激法建立大鼠COPD模型组,其它各药物干预组于制作模型第15天给予芪蛭皱肺无糖颗粒小剂量、中剂量、大剂量和阳性药物溶液灌胃。6周后对COPD模型大鼠肺组织病理切片胶原纤维进行观察,用放免法检测支气管肺泡灌洗液(BALF)以及血清中的层黏连蛋白和透明质酸的含量。结果:与模型组相比,中剂量组、大剂量组、阳性对照组胶原纤维明显减少,胶原平均面积明显减小,BALF以及血清中LN、HA的含量也明显减少;中剂量组胶原平均面积最小优于其它治疗组,大剂量组BALF以及血清中LN、HA的含量较其它治疗组为最少。结论:芪蛭皱肺无糖颗粒,对大鼠模型小气道重塑有重要的干预作用。AIM：To explore the effect of Qizhi Zhoufei Sugar-Free Granules on rat small airway remodeling suf-fered from chronic obstructive pulmonary disease（COPD） by comparing the changes in the level of laminin（LN） and hyaluronic acid（HA） in extra cellular matrix and small airway pathology.METHODS：The rat models of COPD were established by complex stimulation including injection of lipopolysaccharide into intratracheal instilla-tion twice and cigarette smoking inhalation daily.The drug intervention groups were comprised of small dose,mid-dle dose,large dose of Qizhi Zhoufei Sugar-free Granules and positive drug solution on the 15th day.After 6 weeks,the collagenous fibers of rat models lung tissue pathology slice was observed,and the content of LN and HA in bronchoalveolar lavage fluid（BALF） and blood serum were detected using radioimmunoassay method.RE-SULTS：As compared with model group,collagenous fibers in lung the average area of collagen,and the content of LN and HA in BALF and blood serum was significantly decreased in middle dose,high dose and positive drug groups.Middle dose group was better than other treatment groups,in the decrease in average area of collagen,the content of LN and HA in BALF and blood serum in large dose group was the lowest compared with other treatment groups.CONCLUSION：Qizhi Zhoufei Sugar-Free Granules plays an important role in small airway remodeling.
