摘 要:目的:观察血红素加氧酶-1(heme oxygenase-1,HO-1)在高氧暴露早产儿血单个核细胞HO-1蛋白及mRNA的表达水平。方法:分别用反转录聚合酶链反应技术、Western Blot对40例吸入高氧的早产儿(高氧组)血中单个核细胞HO-1 mRNA表达水平及蛋白表达水平进行检测,并与常压空气的早产儿40例对照(对照组)。结果:与对照组相比,高氧组HO-1 mRNA、HO-1蛋白表达明显增加(P<0.001,P<0.01)。结论:高氧暴露可上调早产儿血单个核细胞血红素加氧酶-1的表达,HO-1可能参与了早产儿高氧肺损伤。Objective: To observe the protein and mRNA expression of HO-1 in the peripheral blood mononuclear cell(PBMC) of the hyperoxia-exposed premature infants.Methods: The expressions of HO-1 mRNA and protein in the PBMC of 40 threeday-old hyperoxia-exposed preterm infants(hyperoxia group) and 40 airexposed preterm infants(control group) were measured by reverse transcription polymerase chain reaction(RT-PCR) and Western blot respectivesly.Results: Comparing with the aasay results of the control group,the protein expression of HO-1,mRNA,HO-1 in the hyperoxia group was significantly increased(P〈0.001 and 0.01 respectivesly).Conclusion: Hyperoxia may upregulate the expression of HO-1 in PBMC of hyperoxia-exposed premature infants,and HO-1 may be associated with the hyperoxia-induced lung injury of premature infants.
