摘 要:目的观察七龙天对实验性肺动脉高压(PAH)大鼠内皮素1(ET-1)、白三烯C4(LTC4)的影响。方法雄性SD大鼠随机分为6组:空白对照组(A)、模型组(B)、硝苯吡啶组(C)、七龙天低剂量组(D)、七龙天中剂量组(E)、七龙天高剂量组(F)。空白对照组大鼠以等量0.9%NaCl溶液腹腔注射,其余各组均以野百合碱(MCT)腹腔注射复制PAH模型造模。制模后14天开始,各组分别定时灌胃给药,每日1次,连续给药8天至检查前12h停药。以改良右心导管术测定肺血流动力学参数,用放射免疫(RIA)法测定血浆ET-1,酶联免疫吸附法(ELISA)测定血清LTC4的变化。结果与A组比较,其余各组大鼠PASP、PADP、MPAP、ET-1、LTC4水平较空白对照组均有所升高,且B组升高最显著(P<0.01);与B组比较,C、D、E、F组均可降低大鼠PASP、PADP、MPAP、ET-1水平(P<0.05,P<0.01);与C、D组比较,E、F组大鼠PASP、PADP、MPAP、ET-1水平降低更明显(P<0.05);各用药组对LTC4的影响差异无统计学意义(P>0.05)。结论七龙天能够有效降低实验动物的肺动脉压,其机制与降低ET-1含量有关。Objective To study the effects of "Qilongtian Decoction" on plasma endothelin(ET-1) and leukotrienes C4(LTC4) in pulmonary arterial hypertension(PAH) rats. Methods The PAH model in male SD rats was induced by intraperitoneal injection of monocrotaline(MCT). Nitro-pyridine was used as active control. Parameters of pulmonary hemodynamics were detected by improved right catheterization. Plasma endothelin(ET-1) was detected by radioimmunoassay(RIA). The changes of leukotrienes C4(LTC4) was observed by enzyme-linked immunosorbent assay(ELISA). Results "Qilongtian Decoction" and Nitro-pyridine could significantly reduce the levels of mean pulmonary arterial pressure (MPAP) and ET-1 in PAH rats(P0.05 or P0.01), and little influence the level of LTC4(P0.05); and "Qilongtian Decoction" in middle dose and high dose had better effects than Nitro-pyridine(P0.05). Conclusion "Qilongtian Decoction" can effectively reduce MPAP in PAH rats by reducing ET-1 content.
