摘  要:从快速伸长的纤维细胞中克隆到了一个转录因子,通过序列比对发现其具有典型的MADS-box结构域,命名为GhMADS9。进化树分析表明GhMADS9属于典型的MIKC类MADS转录因子,与在拟南芥胚中高表达的转录因子AGL15亲缘关系最近。RT-PCR和原位杂交试验表明GhMADS9在纤维细胞中特异表达。酵母单杂交试验证明GhMADS9具有转录因子激活活性。继续研究GhMADS9对下游基因的调节机制有利于更加深入地了解棉纤维发育机制。MADs-box genes encode a large family transcription factors which play important roles in diverse developmental processes of flowering plants.GhMADS9 encoding a polypeptide containing a typical MADS-box domain was cloned from 10DPA cotton fibers.Phylogenetic analysis showed that GhMADS9 belonged to MIKCc class of MIKC group,related to AGL15 encoded by the gene which was preferentially expressed in the developing Arabidopsis embryos.RT-PCR and in situ hybridization results indicated that GhMADS9 was expressed specifically in cotton fiber cells.According to the results of yeast one-hybridization,GhMADS9 has transcriptional activity.We suggest that elucidation of GhMADS9 function may eventually provide a new insight into the understanding of cotton fiber development.
