摘  要:选用还原谷胱甘肽作为双功能螯合剂,与牛血清白蛋白偶联,成功螯合了Hg2+,制备汞完全抗原。通过等离子电感耦合法检测抗原与包被原中Hg2+的含量,采用考马斯亮蓝法及SDS-PAGE对抗原进行定性鉴定,并且用TNBS法测定氨基替换率。鉴定完成后用该抗原免疫动物,采用Hg-青霉素-O-VA、青霉素-OVA作包被原检测血清效价,效价大于1∶160000。为制备重金属汞螯合物抗原特异性单克隆抗体提供新途径,为重金属免疫检测技术提供技术依托。In this study,mercury antigen was synthesized using L-Glutathione reduced as the bifunctional chelating agent through the coupling of the bovine serum albumin(BSA) and chelating the mercuryion.The concentration of mercury ion in the antigen and holding antigen was examined with inductively coupled plasma atomic emission spectroscopy(ICPAES).The concentration of protein in the antigen was determined with Bradford method and canned with SDS-PAGE.The degree of conjugate substitution was quantified with trintrobenzene sulfonic acid(TNBS) method.After identifica-tion,the New Zealand white rabbits were immunised with antigen and blood serum was examined with Hg-Penicillin-OVA and Penicillin-OVA as the holding antigens and the titer of rabbit’ blood serum after immunity was greater than 1: 160000.The results indicated that the antigens and holding antigens were synthesized successfully.This research provided a new way for preparing the monoclonal antibody of heavy metal mercury chelate and laid a foundation for setting up the immunity method to exam the heavy metal.
