摘  要:目的筛选大鼠脑创伤3 d后皮层差异表达的miRNA,为颅脑创伤治疗中的miRNA干预策略提供可供选择的目标miRNA。方法将6例创伤后3 d的大鼠大脑皮层组织混匀代表该组样本,将6例正常对照的大鼠大脑皮层组织混匀代表该组样本。利用miRNA芯片检测伤后3 d的创伤大脑皮层组织与正常对照的差异表达基因。并随机挑选差异表达基因作定量聚合酶链反应(PCR)验证。结果与正常对照相比,创伤后3 d组共有251个miRNA表达,其中,2倍以上差异表达上调的20个,2倍以上差异表达下调的5个。定量PCR验证结果提示芯片结果可信。结论创伤大鼠大脑皮层的差异表达miRNA在以前的脑创伤文献中鲜有报道,对其干预有望成为颅脑创伤基因治疗的新方向。Objective This study was designed to screen the different expression of miRNA after traumatic brain injury(TBI) in rats,and to provide the target miRNA for miRNA interference in TBI treatments.Methods Differently expressd genes were examined using miRNA chip in contused cerebral tissue 3days after TBI.The gene that express differently were chose randomly to further be validated by quantitative PCR.Results 251 different expression of miRNA(20 up-regulated and 5 down-regulated more than twice) were found 3days after injury.Quantitative PCR approved the results as well.Conclusion The interference of differently expressed miRNA after TBI would be a new gene thera-py on TBI.
