摘 要:为阐明西瓜G17AB系雄性不育发生的分子机制,采用mRNA差异显示技术,分析了不育株与可育株雄花蕾发育过程中基因表达的差异,经重复验证,获得了2个稳定表达的差异cDNA片段:C13F和G10S。经克隆测序和同源性分析,C13F片段与拟南芥β-葡萄糖苷酶酶蛋白的部分编码序列具有80%同源性;G10S片断与菜豆泛素mRNA的编码序列具有92%同源性。To explore the mechanism of male sterility in watermelon G17AB line, the differences of genetic expression between male sterile buds and male fertile buds in watermelon G17AB line were studied with mRNA differential display method. Two specific cDNA fragments, named as C13F and GIOS ,were obtained. The sequence analysis showed that C13F fragment was 80% identical to code sequence of mRNA of beta-glucosidase-like protein in A rabidopsis thaliana, and G10S fragment was 92% identical to code sequence of Phaseolus vulgaris ubiquitin mRNA. The possible causes of male sterility in watermelon G17AB line were discussed.
