聚乙二醇α-2a对HBV感染患者分泌IL-17变化的研究

摘  要:目的研究应用聚乙二醇α-2a干扰素治疗后发生完全应答的HBV感染患者血清及外周血单核细胞分泌白细胞介素17(IL-17)变化的意义。方法抽取应用聚乙二醇α-2a干扰素治疗发生完全应答HBV感染患者应答点及6个月后及健康对照者、未进行治疗的HBV患者的静脉血,分离血清及单核细胞,检测血清中乙型肝炎病毒表面标志物(HBV-M)、乙型肝炎病毒核酸定量(HBV-DNA)、丙氨酸氨基转移酶(ALT)以及血清中和分别应用抗-CD3、抗-CD28和HBcAb刺激的外周血单核细胞(PBMCs)分泌上清中的IL-17。结果和未进行治疗的HBV患者相比,完全应答者血清中及应用HBcAb刺激的PBMCs分泌的IL-17出现下降;但应用抗-CD3、抗-CD28刺激的出现逐渐升高的趋势,均明显低于正常对照。结论应用聚乙二醇α-2a干扰素治疗有效清除病毒的同时,发生完全应答患者总体分泌IL-17明显下降,有利于减轻炎症反应及肝脏组织的修复。Objective To investigate the change of IL-17 in chronic HBV infection patients who responded completely to pegylated interferon α-2α therapy. Methods We separated the peripheral blood monocytes(PBMCs) and the serum from the blood of HBV infected patients who showed complete response to pegylated interferon α-2α or from those with no therapy and health controls, the PBMCs were cultured with anti-CD3, anti-CD28 or HBcAb together, and the ALT, HBV-DNA and HBV-M of the patients and the IL-17 in the serum and the supematant of different stimuli were detected. Results The IL-17 in the serum and supernatant with HBcAb stimulation in those with complete response was significantly lowered compared with that in those without therapy, but the IL-17 in the supernatant with anti-CD3 and anti-CD28 stimulation was gradually increased. Conclusion Pegylated interferon α-2α therapy can effectively remove HBV, and at the same time, the IL-17 secretion in the patients with complete response was significantly lowered, which will help reduce inflammation and repair liver tissue.
