摘  要:为解决马桑种苗的市场供需矛盾,以马桑茎尖及幼叶为外植体,进行了马桑组织培养技术的初步研究。结果表明,用1%NaCl O消毒20 s加0.1%HgCl消毒15 min,污染率可控制在3%,死亡率控制在6%以下,成活率在91%以上。最佳愈伤组织诱导培养基为MS+0.5 mg/L 6-BA+2.0 mg/L Kt+1.0 mg/L NAA+30g/L蔗糖+8g/L琼脂+0.05%PVP;芽的诱导培养基在MS+6-8g/L琼脂+20-30g/L蔗糖+0.01%-0.05%PVP,pH=5.8-6.0基础上,添加0.5 mg/L 6-BA+0.1 mg/LNAA或1.0 mg/L6-BA+0.2 mg/LNAA;但选择的不同激素浓度配比没有成功诱导出马桑的不定根,使得对马桑不定根的诱导培养成为本研究的难题。The stem tips and young leaves of Coriaria sinica were cultured on different callus and bud induction media to screen the suitable medium for its callus and bud induction.The results showed that the contamination rate,death rate and survival rate of explants sterilized by 1%NaClO for 20 s and 0.1%HgCl for 15 s were 3%,below 6% and above 91% respectively,the optimum medium for callus induction was MS+0.5 mg/L 6-BA +2.0 mg/L Kt +1.0 mg/L NAA +30 g/L sucrose +8 g/Lagar + 0.05% PVP and the optimum media for bud induction were MS+0.5 mg/L 6-BA+0.1 mg/LNAA + 6~8 g/L agar+20~30 g/L sucrose+0.01%~0.05% PVP and MS+1.0 mg/L 6-BA+0.2 mg/L NAA + 6~8 g/L agar+20~30 g/L sucrose+0.01%~0.05% PVP.However,adventitious root of Coriaria sinica was not induced on the media with different hormone concentration which holding back the research.
