摘  要:目的考察黄芪甲苷(AS-IV)对迟发型超敏反应的影响。方法应用2,4-二硝基氯苯(DNCB)造成小鼠变态反应性接触性皮炎(ACD)模型,分别考察(AS-IV)诱导相和效应相给药对耳廓肿胀的影响,并进行耳病理组织学检查;ELISA方法检测小鼠血清IFN-γ水平。MTT法检测ConA诱导的脾细胞增殖。结果 (AS-IV)诱导相给药可明显抑制ACD小鼠耳肿胀,而效应相给药未观察到明显效应;病理组织学结果显示(AS-IV)诱导相给药可减轻模型小鼠局部水肿及血管扩张,减少淋巴细胞浸润;(AS-IV)诱导相给药明显降低血清IFN-γ水平;离体实验未发现(AS-IV)对ConA诱导的脾细胞增殖有明显影响。结论 (AS-IV)诱导相给药有抑制迟发型超敏反应的作用,该作用可能与抑制IFN-γ有关。OBJECTIVE To evaluate the effects of astragaioside Ⅳ{AS-Ⅳ) on delayed type hypersensitivity (DTH). METHOD Murine allergic contact dermatitis (ACD) models were induced by DNCB in AS-Ⅳ induction phase and reaction phase to observe their effects and conduct the auricle histopathological examination respectively. Mice IFN-γ in serum was detected by ELISA. Splenocytic proliferation induced by ConA was measured by TTA. RESULT AS-IV induction phase evidently inhibited the auricle inflammation of ACD models; however, effector phase had no obvious effect. Hestopathological results indicated that AS-Ⅳ induction phase alleviated local edema and angiectasis of mice models and reduced lymphocytic infiltration. AS-Ⅳ induction phase markedly decreased IFN-γ levels in serum. And in vitro study, AS- Ⅳ had no obvious effect on proliferation of splenocytes induced by ConA. CONCLUSION AS-Ⅳ induction phase can restrain DTH ,which may be due to its inhibition to IFN-γ.
