摘　　要：目的建立简便可靠的检测抗-Jo-1抗体的方法。方法用丙酮沉淀兔胸腺中的总蛋白制成兔胸腺丙酮粉,从兔胸腺丙酮粉中提取可提取性核抗原(ENA),ENA经亲和色谱纯化出Jo-1抗原,将纯化的Jo-1抗原制备成斑点印迹(DB)试剂盒,用该试剂盒检测多种风湿免疫性疾病和正常血清中的抗-Jo-1抗体,探讨各种血清中抗-Jo-1抗体的阳性率。结果 DB试剂盒共检测92份血清中的抗-Jo-1抗体。各种风湿性疾病血清69份,其中多发性肌炎(DM)20份,阳性6份(30%);其他风湿性疾病49份,无阳性。健康对照23份,无阳性。该试剂盒对诊断DM的敏感度为30%,特异度为100%。结论 DB试剂盒检测抗-Jo-1抗体方法简便,结果可靠,具有较大的临床开发应用价值。Objective To develop a convenient and reliable assay for detecting anti-Jo-1 antibody.Method The total protein was precipitated from rabbit thymus by acetone,and then was made into rabbit thymus acetone powder.The ENA was extracted from rabbit thymus acetone powder.The Jo-1 antigen was purified from the ENA by immunoaffinity chromatography.The purified Jo-1 antigen was made into DB kit.The anti-Jo-1 antibodies of various rheumatic disease and normal control sera were detected by the kit.Results The anti-Jo-1 antibodies of 92 sera samples were detected by the DB kit.There were 20 samples sera of DM and 49 samples sera of other rheumatic diseases.The anti-Jo-1 antibodies were detected in 6（30%） samples of DM and 0 sample of other rheumatic diseases,respectively.Among 23 control sera,there was no anti-Jo-1 antibody by the kit.To diagnose DM by the kit,the sensitivity and specificity were 30% and 100%,respectively.Conclusion The anti-Jo-1 antibodies detected by the DB kit was convenient and reliable.It might be a promising assay for detecting anti-Jo-1 antibodies in clinical laboratory.
