经血宁胶囊5种提取部位凝血作用的药效筛选

摘　　要：目的比较经血宁胶囊5种提取部位之间凝血作用的差异,明确其凝血作用部位。方法对经血宁胶囊5种提取部位即石油醚部位(A)、乙酸乙酯部位(B)、正丁醇部位(C)、水溶部位(D)、残渣部位(E)进行凝血作用的药效筛选。(1)采用毛细玻管法和玻片法测定凝血时间(coagulation time,CT),观察经血宁胶囊5种提取部位对小鼠凝血时间的影响;(2)凝血酶原时间(prothrombin time,PT)测定;(3)活化部分凝血活酶时间(activated partial thromboplastin time,APTT)测定,探讨其凝血作用机制。结果 (1)毛细玻管法和玻片法测定小鼠凝血时间(重复3次),结果显示经血宁胶囊5种提取部位在9 g生药/kg剂量下均有不同程度的缩短凝血时间,其中正丁醇部位(C)重现性较好,提示正丁醇部位(C)具有较好的凝血作用。A、D、E提取部位凝血作用不明显。(2)经血宁胶囊5种提取部位的家兔PT时间分别与空白对照组比较,正丁醇部位(C)P<0.03,残渣部位(E)P<0.01,PTR值较小,提示C、E部位具有较好的缩短凝血酶原(PT)时间作用,其他部位影响凝血酶原时间不明显。(3)经血宁胶囊5种提取部位的血浆APTT分别与空白对照组比较,A、C、D、E部位均有显著性差异(P<0.01),提示均有具有较好的缩短APTT作用。结论经血宁胶囊正丁醇部位(C)在9 g生药/kg剂量下具有较好的缩短凝血时间的作用,且重复性较好,其他提取部位凝血作用不明显。Objective To observe and compare the hemagglutination of five extract parts of Jingxuening capsule,i.e.,petroleum ether（A）,ethyl acetate extract（B）,n-butanol extract（C）,water extract（D）,and residue extract（E）,and to find the hemagglutination part of Jingxuening Capsule.Methods（1） Glass capillary method and slide method were used to examine the coagulation time（CT） for the observation of the effect of five extract parts of Jingxuening Capsule on coagulation time of mice.（2） Prothrombin time（PT） was determined.（3） Activated partial thromboplastin time（APTT） was determined to explore its hemagglutination mechanism.Results（1）The results of determination by glass capillary method and slide method（triplicate） showed that the five extract parts of Jingxuening Capsule（9 g/kg） shortened CT to different degrees,and n-butanol extract had good repeatability,which suggested that n-butanol extract had better hemagglutination,but hemagglutination was not obvious in parts A,D and E.（2） Compared with the blank control group,prothrombin ratio was lower in n-butanol extract（C）（P〈0.03） and in leaving extract（E）（P〈0.01）,which suggest that parts C and E could significantly shorten PT,while others had no obvious effect on PT.（3） Compared with the blank control group,APTT had significant difference in parts A,B,C,D,and E（P0.01）,which suggest that all of the extract parts could significantly shorten the APTT.Conclusion n-butanol extract of Jingxuening Capsule（9 g/kg） has notable effect of shortening coagulation time,and is with a good repeatability,while hemagglutination is not obvious in other parts.
