摘  要:目的研究比较3种五环三萜化合物齐墩果酸(OA)、乌苏酸(UA)、积雪草酸(AA)的护肝及抗氧化作用。方法建立小鼠D-氨基半乳糖/脂多糖(D-GalN/LPS)肝损伤模型,检测OA、UA、AA对损伤小鼠血清天冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)活性以及肝组织脂质过氧化的影响;运用体外自由基清除试验,测定3种化合物对超氧阴离子和羟自由基的清除作用。结果 100 mg.kg-1OA、UA、AA均能有效抑制D-GalN/LPS引起的小鼠血清AST及ALT活性的显著上升;明显减少肝组织脂质过氧化产物MDA的生成;一定浓度3种化合物能有效清除超氧阴离子和羟自由基,且3个化合物清除自由基能力存在显著差异。结论 3种五环三萜化合物OA、UA、AA均具有显著护肝和抗氧化活性,且AA、UA的护肝作用优于OA,其机理可能与其具有更强的清除自由基能力有关。Objective To compare the hepatoprotective and anti-oxidative effects of three pentacyclic triterpenoids,oleanolic acid(OA),ursolic acid(UA) and asiatic acid(AA).Methods D-GalN/LPS-induced liver injury model was used to evaluate the effects of OA,UA and AA on the activities of serum aspartate aminotransferase(AST) and alanine aminotransferase(ALT) and the level of liver lipid peroxidation in mice.Scavenging activity on superoxide anion and hydroxyl radicals of three compounds were investigated to determine their anti-oxidative effects.Results Pretreatment with 100 mg·kg-1 OA,UA and AA,dovusly reversed the increase of serum AST and ALT activities and the level of liver lipid peroxidation induced by D-GalN/LPS in mice.OA,UA and AA,at concentrations ranging from 50 to 450 mol·L-1,showed dose-dependent superoxide anion and hydroxyl radical scavenging activity.In addition,it was also found that there were differences among three compounds in scavenging activity on free radicals,anti-oxidative and hepatoprotective effects.Conclusion Three pentacyclic triterpenoids,OA,UA and AA,have significant hepatoprotective activity against D-GalN/LPS-induced liver injury in mice.Moreover,the hepatoprotection and anti-oxidation of UA and AA are stronger than that of OA and the mechanisms might be related to their more powerful scavenging activities on free radicals.
