白花夹竹桃愈伤组织的诱导试验

摘　　要：[目的]探讨夹竹桃愈伤组织诱导的适宜条件,为更好开发夹竹桃的药用价值奠定基础。[方法]用不同的消毒方法和激素配比对夹竹桃的叶片和茎段进离体培养,诱导愈伤组织。[结果]对夹竹桃外植体采用自来水冲洗1h,75%酒精处理30s,4%NaClO消毒12～16min能取得较好的消毒效果;茎段更适宜做为诱导愈伤组织的外植体,在MS培养基加0.05mg/L2,4-D和0.15mg/L6-BA的条件下,能高效率的诱导出愈伤,且生长较好;[结论]该研究为进一步筛选产强心苷的高产细胞系奠定了基础。[Objective] To explore the optimum induction conditions of oleander callus for the development of the oleander medicinal value.[Method] Oleander leaf and stem callus were cultured using different disinfection methods and different hormone in vitro.[Result] The results show that the oleander explants used by water washing 1 h,75% ethanol for 30 s,4% NaClO sterilized 12-16 min can get better disinfection,and stems are more suitable as explants for callus.In the condition of MS basic medium added with 0.05 mg /L 2,4-D and 0.15 mg/L 6-BA,the callus developed better.[Conclusion] This research lays foundation for further screening production lines of cardiac glycosides.
