摘  要:目的:研究SERCA2a mRNA在左心室机械辅助减负荷模型中的表达及其意义,探讨左心室减负荷后心肌分子水平的变化。方法:选用Lewis大鼠,结扎冠状动脉左前降支诱导心力衰竭4周后,移植心力衰竭后的心脏及右肺到受体大鼠腹部(通过供体的升主动脉与受体的降主动脉吻合)。移植2周后评价心脏质量,左心室质量,心肌细胞直径,SERCA2a mRNA的表达情况。结果:心力衰竭后增大的心脏左心室质量、左心室心肌细胞直径于移植后趋于正常;心力衰竭后左心室SERCA2a表达明显降低,移植后趋于正常。结论:左心室减负荷后,心肌逆向重塑伴随着心肌细胞分子水平改变,可能与心肌收缩功能的恢复有关。Objective To study the expression and significance of sarcoplasmic reticulum Ca2+ATPase mRNA in the model of left ventricular mechanical unloading and to explore the change of cardiomyocyte in molecular level.Methods Heart failure was induced in Lewis rats by ligating left anterior descending artery.After four weeks,the heart and right lung were heterotopically transplanted into the abdomen of the recipients by anatomizing their ascending aorta to the recipients' abdominal aorta.Two weeks after transplantation,cardiac mass,left ventricular mass,diameter of myocardial cell and the expression of sarcoplasmic reticulum Ca2+ATPase were evaluated.Results The increased left ventricular mass and diameter of myocardial cell became normal after transplantation.The expression of sarcoplasmic reticulum Ca2+ATPase was obviously lowered in the failure heart and turned normal after transplantation.Conclusion The myocardium reverse remodeling after left ventricular unloading is accompanied by the change of cardiomyocyte in molecular level.And the change in molecular level may be related to the improvement of myocardium contractive function.
