摘 要:目的利用原子力显微镜技术(AFM)和荧光技术研究原代培养的海马神经元间的连接结构及其相关功能。方法选择生长良好的海马神经元,用2.5%的戊二醛(生理盐水配制)固定30 min,置于AFM的扫描器上进行扫描成像。选择生长良好取自转基因RFP C57乳小鼠、EGFP C57乳小鼠(其脑部特异性地转入绿色荧光蛋白和红色荧光蛋白)的原代混合共培养海马神经元细胞,在荧光显微镜下观察并拍照。结果与结论正常海马神经元表面光滑,起伏均匀、规律,突起及细胞之间的超微结构清晰,神经元胞体间存在膜性连接;在共培养的细胞中,发现两个通过膜性连接的海马神经元中的一个细胞内,同时存在红色和绿色荧光蛋白。神经元之间存在膜性,且膜性连接结构可能存在大分子的物质交换或转运。Objective To investigate the connection between primarily cultured hippocampal neurons using atomic force microscopy(AFM),and to explore the molecular communication or material transportation between primarily cultured hippocampal neurons.Methods Well-growing hippocampal neurons chosen under the light microscope were fixed in 2.5% glutaraldehyde of normal saline solution for 30 min.The culture dish with the interesting structures was immobilized onto the base of AFM,which was then put onto the top of the AFM scanner.Well-growing hippocampal neurons from the co-cultured RFP C57 new-born mice and EGFP C57 new-born mice were observed by the fluorescence microscope.Results Hippocampal neurons displayed a smooth membrane surface with even and regular undulation.There were continuous membranous junctions(CMJ) between two closely adjacent neurons,and there were both green fluorescent protein and red fluorescent protein in one of the two closely adjacent neurons connected to the CMJ.Conclusion CMJs between two closely adjacent neurons are found,through which there may be some molecular communication or material transportation.
