摘 要:[目的]通过纤维环穿刺法建立兔椎间盘退变模型,分析其特点。[方法]将16只新西兰大白兔随机分为2组。经腹膜外入路暴露L2、3、L3、4椎间隙。A组为纤维环穿刺组,采用16号针头穿刺,B组为假手术对照组。术后2、4、6、8周通过MR I和组织病理学检查观察髓核变性及组织病理情况。[结果]纤维环穿刺组髓核信号强度在术后呈现逐渐降低趋势,髓核面积逐渐缩小,椎间隙高度也逐步下降,从术后4周开始两组差异有统计学意义(P<0.05)。随着时间的进展,纤维环穿刺组髓核内细胞含量逐渐减少。[结论]纤维环穿刺法可成功建立椎间盘退变模型,比较真实地模拟了人类椎间盘损伤后的退变过程。[Objective]To construct the rabbit model of degenerative intervertebral disc by anulus puncture and analyze its characteristics. [Methods]Sixteen New Zealand white rabbits were randomly divided into 2 groups.Intervertebral discs of L2、3 and L3、4 were exposed by extro-peritoneal approach.Group A was for annulus puncture with an 16-gauge needle and group B was used as a control.Pathological change of intervertebral disc was checked with MRI and histopathological examination at 2,4,6,and 8 weeks after operation.[Results]After operation in Group A,the intensity of nucleus gelatinosus was declined,the area of nucleus gelatinosus was deflated and the height of intervertebral space was also decreased,but the significant changes occurred from the fourth week on(P0.05).The histological sections demonstrated that the cell content in nucleus pulposus was gradually increased.[Conclusion]The rabbit model of intervertebral disc degeneration can be successfully constructed by the method of anulus puncture.This method can authentically imitate intervertebral disc degeneration in human beings.
