摘 要:目的:研究江南牡丹草总生物碱对大白鼠佐剂关节炎的继发性病变的治疗效果。方法:本实验采用建造大白鼠佐剂关节炎模型的方法,观察不同浓度的药物对大白鼠佐剂关节炎继发病变的治疗作用,36只大白鼠按性别随机分为高剂量组(200mg/kg)、低剂量组(100mg/kg)和正常对照组(每组雌雄各半),测量大白鼠局部肢体的体积,并观察其一般情况,观察各给药组对大白鼠佐剂关节炎的治疗效果;另取108只大白鼠分为正常对照组、致炎组和高剂量组(200mg/kg)用于血沉测定,通过测定不同时期血沉的变化,间接反映药物的治疗效果。结果:江南牡丹草总生物碱对大白鼠佐剂关节炎引起的继发性局部肢体肿胀有一定的消肿作用。江南牡丹草总生物碱高剂量组第18天、第24天、第32天对大白鼠的佐剂关节炎继发性病变引起的肿胀有一定的治疗效果(与正常对照组相比,P<0.05),并且其对大白鼠的一般情况没有明显的异常影响,而江南牡丹草总生物碱低剂量组对于大白鼠佐剂关节炎的继发性病变未见明显作用(与正常对照组相比,P>0.05);致炎后血沉数值显著升高,而用药后血沉数值逐渐降低(与致炎组相比差异有统计学意义P<0.05)。结论:江南牡丹草总生物碱高剂量给予患佐剂关节炎的大白鼠,能够显著降低血沉及其肢体的继发性肿胀,对其继发性病变有一定的治疗效果。Objective:To explore the effect of Leontice kiangnanensis total alkaloids (LKTA) on adjuvant arthritis in rats. Methods:After adjuvant arthritis models were established, thirty-six rats are divided into high dose group (200mg/kg), low dose group (100mg/kg), and normal control group. Before observing general conditions and the treatment effect on adjuvant arthritis in each treatment group, we measured the volume of the local limb in rats. Another 108 rats were divided into normal control group, inflammation-causing group and high dose group (200mg/kg) for observing erythrocyte sedimentation rate (ESR) which can show the treatment effect of LKTA indirectly. Results:LKTA had certain detumescence effects on secondary local limb swelling caused by adjuvant arthritis in rats. High dose group of LKTA presented certain treatment effect on swelling caused by adjuvant arthritis on the 18th, 24th, and 32th (compared with control group, P0.05). Nevertheless, there was little toxicity on rat in general condition. However, low dose group of LKTA showed little treatment effect on swelling (compared with control group, P0.05).The values of ESR increased obviously after causing inflammation, while decreased gradually after administration (compared with inflammation-causing group, P0.05). Conclusion:High dose of LKTA is able to significantly reduce the value of ESR and the secondary limb swelling. It has certain treatment effect on secondary lesion in rats with adjuvant arthritis.
